In addition to their role in cutting DNA molecules, restriction enzymes help in the initial steps of analysing nucleotide sequences. This exercise demonstrates the principles of restriction site mapping, using DNA from the lambda bacteriophage. Two or three enzymes can be used to cut the DNA, as desired. A 1 kb 'DNA ladder' provides a 'molecular ruler' with fragments of known sizes against which the lambda fragments can be measured. It is then possible to deduce the relative positions of the restriction sites of the two or three enzymes on the lambda DNA. | ||||||
